Asu no Yoichi! bercerita tentang Yoichi Karasuma telah menghabiskan seluruh hidupnya di pegunungan, berlatih dengan aliran pedan Siulan Angin. Di bawah bimbingan ayahnya, dia mampu menguasai semua teknik itu pada usia 17. Dengan tidak ada yang tersisa untuk dipelajari, Yoichi dikirim ke sebuah dojo baru yang terletak di kota sehingga dia dapat terus melatih dan mendapatkan pemahaman tentang masyarakat modern.
Sayangnya, Yoichi tidak tahu bagaimana harus bertindak atau berbicara kepada siapapun di hari ini dan bertindak seperti samurai, lengkap dengan ucapan aneh dan pakaian tradisional. Saat dia pergi untuk tinggal dengan saudara perempuan Ikaruga di dojo, Yoichi, tidak mengerti bagaimana berinteraksi dengan orang lain, terus-menerus terdorong oleh kesalahpahaman yang lucu. Asu tidak Yoichi! Mengikuti Yoichi saat ia tersandung melalui kehidupan barunya dan mencoba untuk belajar bagaimana hidup di dunia modern dengan keluarga barunya.
